ABSTRACT

BACKGROUND Panstrongylus rufotuberculatus (Hemiptera-Reduviidae) is a triatomine species with a wide geographic distribution and a broad phenotypic variability. In some countries, this species is found infesting and colonising domiciliary ecotopes representing an epidemiological risk factor as a vector of Trypanosoma cruzi, etiological agent of Chagas disease. In spite of this, little is known about P. rufotuberculatus genetic diversity. METHODS Cytogenetic studies and DNA sequence analyses of one nuclear (ITS-2) and two mitochondrial DNA sequences (cyt b and coI) were carried out in P. rufotuberculatus individuals collected in Bolivia, Colombia, Ecuador and Mexico. Moreover, a geometric morphometrics study was applied to Bolivian, Colombian, Ecuadorian and French Guiana samples. OBJECTIVES To explore the genetic and phenetic diversity of P. rufotuberculatus from different countries, combining chromosomal studies, DNA sequence analyses and geometric morphometric comparisons. FINDINGS We found two chromosomal groups differentiated by the number of X chromosomes and the chromosomal position of the ribosomal DNA clusters. In concordance, two main morphometric profiles were detected, clearly separating the Bolivian sample from the other ones. Phylogenetic DNA analyses showed that both chromosomal groups were closely related to each other and clearly separated from the remaining Panstrongylus species. High nucleotide divergence of cyt b and coI fragments were observed among P. rufotuberculatus samples from Bolivia, Colombia, Ecuador and Mexico (Kimura 2-parameter distances higher than 9%). MAIN CONCLUSIONS Chromosomal and molecular analyses supported that the two chromosomal groups could represent different closely related species. We propose that Bolivian individuals constitute a new Panstrongylus species, being necessary a detailed morphological study for its formal description. The clear morphometric discrimination based on the wing venation pattern suggests such morphological description might be conclusive.

ABSTRACT

Gymnorhamphichthys britskii is a Neotropical electric fish of family Rhamphichthyidae described from the Paraná-Paraguay system. This study reports the first karyotypic description of G. britskii collected from the upper Paraná river basin, which presented 2n=38 chromosomes, karyotype composed of 14 metacentric, 8 submetacentric, 2 subtelocentric and 14 acrocentric chromosomes, and fundamental number as 62 for both sexes. Heteromorphic sex chromosomes were absent. A single pair of nucleolar organizing regions (NORs) was detected in the submetacentric chromosome pair number 9 by silver staining and confirmed by the 18S rDNA probe. The 5S rDNA was located in a single chromosome pair. Heterochromatic regions were clearly observed in the short arms of the NOR-bearing chromosome pair and in the telomeric positions of most acrocentric chromosomes. Besides the present data are valuable to help in understanding karyotypic evolution in Rhamphichthyidae, data from NORs confirmed the tendency of this family in presenting simple NORs sites, similar to the other Gymnotiformes clades. Yet, the presence of a large heterochromatic block in the NOR-bearing chromosome can be used as cytogenetic markers for G. britskii, and that centric fusions appear to be an important mechanism in the karyotype evolution and differentiation among Gymnotiformes species.(AU)


Gymnorhamphichthys britskii é um peixe neotropical da família Rhamphichthyidae descrita no sistema Paraná-Paraguai. Este estudo relata a primeira descrição cariotípica de G. britskii coletado na bacia do alto rio Paraná, que apresentou 2n = 38 cromossomos, cariótipo composto por 14 metacêntricos, 8 submetacêntricos, 2 subtelocêntricos e 14 acrocêntricos, e número fundamental 62 para ambos sexos. Cromossomos sexuais heteromórficos estavam ausentes. Um único par de regiões organizadoras de nucléolos (RONs) foi detectado no par de cromossomos submetacêntricos número 9 por coloração com prata e confirmado pela sonda DNAr 18S. O DNAr 5S foi localizado em um único par cromossômico. Regiões heterocromáticas foram claramente observadas nos braços curtos do par de cromossomos que carrega a RON e nas posições teloméricas da maioria dos cromossomos acrocêntricos. Além dos dados presentes serem valiosos para auxiliar na compreensão da evolução cariotípica em Rhamphichthyidae, dados de RONs confirmaram a tendência desta família em apresentar sítios simples de RONs, semelhantes aos demais clados de Gymnotiformes. No entanto, a presença de um grande bloco heterocromático no cromossomo portador da RON, pode ser usado como marcador citogenético para G. britskii e as fusões cêntricas parecem ser um mecanismo importante na evolução e diferenciação cariotípica entre as espécies de Gymnotiformes.(AU)

ABSTRACT The karyotypes of three armored catfish species (Loricariidae) from the Iguaçu river, southern of the Brazil, were compared using different techniques: C-banding, Ag-NOR and fluorescence in situ hybridization (FISH), which used 5S and 18S rDNAs and total Cot-1 fraction as probes. Hypostomus commersoni and Hypostomus derbyi presented 2n = 68 chromosomes, with karyotype formulae 12m+12sm+14st+30a and 12m+12sm+10st+34a, respectively; whereas Hypostomus myersi presented 2n = 74 chromosomes and 12m+16sm+12st+34a. The chromosomal localization of the Ag-NORs, 5S and 18S rDNAs differed in number of sites and chromosomal localization among the studied species. The total Cot-1 probe permitted the visualization of the repetitive DNA fraction in karyotypes of each species. Cross-hybridizations using total Cot-1 probe revealed that these species have repetitive DNAs in common. However, this does not occur in H. commersoni in relation to the other species. The apparent karyotype similarity suggests a close relationship between the sympatric H. commersoni and H. derbyi species, but the small differences detected in the examined chromosomal markers indicate evolutionary divergence due to gene flow restriction among them. Hence, the present findings indicate different composition of repetitive sequences among studied species, which permit to infer its role in chromosomal differentiation of Hypostomus.

ABSTRACT

The fish species Synbranchus marmoratus has been reported to exist as a species complex due to high intraspecific karyotypic variability in spite of the difficulty or impossibility to distinguish them using morphological traits alone. The goal of this work was to use cytogenetic and molecular methods to determine the species delimitations and understand the karyoevolution of S. marmoratus using samples collected from distinct Brazilian localities. Among the analyzed specimens, a large degree of cytogenetic variation related to diploid numbers and karyotype structure was observed, with karyotypes showing 2n=42, 44 and 46 chromosomes. In addition, using sequences of three mitochondrial genes, the phylogenetic relationships between every sample with a known karyotype were determined, which revealed significant nucleotide divergence among the karyomorphs. Also, the analyses indicate that chromosomal rearrangements occurred independently within the distinct lineages of S. marmoratus complex, which resulted in the appearance of distinct karyotypic variants in a non-linear fashion related to diploid numbers and in the appearance of similar non-homologous chromosomes. Finally, the integration of both molecular cytogenetic and phylogenetic approaches allowed the determination of specific chromosomes possibly involved in rearrangements and a better understanding about the evolutionary processes involved in the differentiation of Synbranchus genus.


A espécie de peixe Synbranchus marmoratus tem sido reportada como um complexo de espécies devido à elevada variabilidade cariotípica intraespecífica a despeito da dificuldade ou impossibilidade de distingui-las usando apenas caracteres morfológicos. O objetivo deste trabalho foi utilizar métodos citogenéticos e moleculares para determinar a delimitação das espécies e compreender a carioevolução de S. marmoratus utilizando amostras coletadas em distintas localidades brasileiras. Dentre os espécimes analisados, um alto grau de variação citogenética relativo aos números diploides e estrutura cariotípica foi observado, com cariótipos mostrando 2n=42, 44 e 46 cromossomos. Adicionalmente, utilizando sequências de três genes mitocondriais, as relações filogenéticas entre cada amostra com cariótipo conhecido foram determinadas, revelando uma divergência nucleotídica significativa entre os cariomorfos. Além disso, as análises indicam que rearranjos cromossômicos ocorreram independentemente nas distintas linhagens do complexo S. marmoratus, o que resultou no aparecimento de distintas variantes cariotípicas de forma não linear em relação aos números diploides e no surgimento de cromossomos similares e não homólogos. Finalmente, a integração de uma abordagem citogenética molecular e filogenética permitiu a determinação de cromossomos específicos que, possivelmente, estão envolvidos em rearranjos e um melhor entendimento sobre os processos evolutivos envolvidos na diferenciação do gênero Synbranchus.

ABSTRACT

The lacustrine system of the middle rio Doce basin is considered a paradigm of Pleistocene geomorphology. In these lakes, two Hoplias malabaricus karyomorphs (2n = 42A and 2n = 42B) live in sintopy in Carioca Lake. Cytogenetic analyses were performed on 65 specimens from 8 lakes (including Carioca Lake) to determine the distribution and relative frequency of these karyomorphs and the degree of cytogenetic divergence caused putatively by recent geographic isolation. All fish were 2n = 42B karyomorphs, except for 1 specimen from the Marola Lake, which was 2n = 42A. Among-population variation was especially high for C-banding patterns. Other characters such as X chromosome size and CMA3/DAPI also varied among populations. Our results suggested that the karyotype of H. malabaricus is able to respond rapidly to geographic isolation, and revealed that heterochromatic variation may represent the lowest hierarchical level of chromosomal evolution.


O sistema lacustre da bacia do médio rio Doce é considerado um paradigma da geomorfologia do Pleistoceno. Nestes lagos, dois cariomorfos de Hoplias malabaricus (2n = 42A e 2n = 42B) vivem em sintopia na lagoa Carioca. Análises citogenéticas foram realizadas em 65 amostras de 8 lagos (incluindo lagoa Carioca) para determinar a distribuição e frequência relativa destes cariomorfos e o grau de divergência citogenética aparentemente causada pelo isolamento geográfico recente. Todos os peixes apresentaram o cariomorfo 2n = 42B, com exceção de 1 espécime da lagoa Marola, que foi 2n = 42A. Entre as populações, a variação foi especialmente elevada nos padrões de bandamento C. Outros caracteres como o tamanho do cromossomo X e os padrões de CMA3/DAPI também variaram entre as populações. Nossos resultados sugerem que o genoma de H. malabaricus é capaz de responder rapidamente ao isolamento geográfico, revelando que a variação de heterocromatina pode representar o nível hierárquico mais baixo de evolução cromossômica.

ABSTRACT

Juliomys is a small rodent from the family Cricetidae which inhabits the Atlantic forest and forests from Argentina to eastern Brazil. The three species recognized so far have different karyotypes. In this paper, we describe a new karyotype with 2n = 32, FN = 48 found in Juliomys specimens from a high-altitude area in the Atlantic forest of southern Brazil. The karyotype was analyzed after G- and C-banding and silver staining of the nucleolus organizer regions (Ag-NOR) and its G-banding patterns were compared with those of the newly described species Juliomys ossitenuis (2n = 20, FN = 36). The 2n = 32 karyomorph presented peculiar features and was very different from those of the other species of the genus: J. pictipes (2n = 36, FN = 34), J. rimofrons (2n = 20, FN = 34) and J. ossitenuis (2n = 20, FN = 36). Differences were mostly due to centric and tandem fusions, pericentric inversion and loss of heterochromatin. The karyotype represents a powerful tool to differentiate Juliomys species and our data suggest that the karyotype described herein belongs to a new species.

Pseudacteon flies, parasitoids of worker ants, are being intensively studied as potentially effective agents in the biological control of the invasive pest fire ant genus Solenopsis (Hymenoptera: Formicidae). This is the first attempt to describe the karyotype of P. curvatus Borgmeier, P. nocens Borgmeier and P. tricuspis Borgmeier. The three species possess 2n = 6; chromosomes I and II were metacentric in the three species, but chromosome pair III was subtelocentric in P. curvatus and P. tricuspis, and telocentric in P. nocens. All three species possess a C positive band in chromosome II, lack C positive heterochromatin on chromosome I, and are mostly differentiated with respect to chromosome III. P. curvatus and P. tricuspis possess a C positive band, but at different locations, whereas this band is absent in P. nocens. Heterochromatic bands are neither AT nor GC rich as revealed by fluorescent banding. In situ hybridization with an 18S rDNA probe revealed a signal on chromosome II in a similar location to the C positive band in the three species. The apparent lack of morphologically distinct sex chromosomes is consistent with proposals of environmental sex determination in the genus. Small differences detected in chromosome length and morphology suggests that chromosomes have been highly conserved during the evolutionary radiation of Pseudacteon. Possible mechanisms of karyotype evolution in the three species are suggested.

ABSTRACT

Hypostomus is the most speciose genus in the family Loricariidae, with approximately 120 species. These fish show a wide morphological and color variation, which hinders the identification of species, mainly of widely distributed representatives. The aim of this study was to contribute to the current knowledge on cytogenetic features of Hypostomus nigromaculatus. Three specimens of H. nigromaculatus, collected in two tributaries of rio Tibagi, Paraná, and in Cachoeira de Emas, rio Mogi-Guaçu, São Paulo, the latter being the type locality of H. nigromaculatus, were studied. Chromosomal preparations were submitted to Giemsa staining, silver nitrate impregnation, C-banding and CMA3 and DAPI fluorochromes staining. All samples presented 2n = 76, but the rio Mogi-Guaçu sample differed from those from tributaries of rio Tibagi in relation to karyotype formulae, distribution and composition of heterochromatin, and NOR location. The silver nitrate staining revealed the presence of multiple Ag-NORs for all samples, but with differences on the location on chromosomes. CMA3 staining reveled bright signals equivalent to NOR-bearing chromosomal segments; such sites were characterized by negative, i.e. unstained, marks after DAPI staining. The pattern of heterochromatin distribution was distinctive among samples from rio Mogi-Guaçu and tributaries of rio Tibagi. The differences observed between the sample from rio Mogi-Guaçu and the ones from tributaries of rio Tibagi allow us to suggest that these samples are presently isolated. Further analyses are necessary to ascertain whether such isolation refers to distinct populations or characterizes true different species.


O gênero Hypostomus é um dos mais especiosos na família Loricariidae, tendo aproximadamente 120 espécies. Apresenta uma ampla diversidade quanto ao padrão de coloração e morfologia, o que dificulta a identificação de determinadas espécies, principalmente aquelas com ampla distribuição geográfica. Para isso os dados obtidos neste trabalho contribuem para os estudos citogenéticos de Hypostomus nigromaculatus. Foram analisados três exemplares de H. nigromaculatus de afluentes do rio Tibagi, Paraná e Cachoeira de Emas, rio Mogi-Guaçu, São Paulo, sendo esta última, a localidade tipo de H. nigromaculatus. Os cromossomos foram submetidos à coloração convencional (Giemsa), impregnação por nitrato de prata, bandamento-C e coloração com os fluorocromos CMA3 e DAPI. Todos os exemplares apresentaram 2n = 76, no entanto com diferença quanto às fórmulas cariotípicas, distribuição e composição da heterocromatina. O nitrato de prata detectou RONs múltiplas para as amostras, porém com diferenças quanto à localização nos cromossomos. A coloração com fluorocromo CMA3 foi correspondente aos cromossomos Ag-RONs, na coloração com DAPI foram observadas bandas negativas, ou seja, não coradas. O padrão de distribuição da heterocromatina foi diferente para as amostras do rio Mogi-Guaçu e dos tributários do rio Tibagi. As diferenças observadas entre as amostras de rio Mogi-Guaçu e afluentes do rio Tibagi permitem-nos sugerir que essas amostras estejam atualmente isoladas. Outras análises são necessárias para determinar se as amostras analisadas são populações distintas ou trata-se de espécies diferentes.

Karyotype data are presented for distinct species of the genus Astyanax from four rivers belonging to three different hydrographic basins of the State of Paraná, Brazil (Verde River - Tibagi basin, Açungui River - Ribeira basin, and Santo Antônio and Jaguariaíva Rivers - Jaguariaíva basin). Three karyotypic forms were identified, here denominated karyotype A (2n = 50 chromosomes, with 8m+18sm+10st+14a, and thirteen 18S rDNA sites); karyotype B (2n = 50 chromosomes, with 8m+18sm+10st+1f4a, and four 18S rDNA sites); and karyotype C (2n = 48 chromosomes, with 10m+16sm+10st+12a, and eight 18S rDNA sites). The pattern of constitutive heterochromatin was similar among the three karyotypic forms, with few differences. The 5S rDNA corresponds to a synapomorphic character regarding its number and chromosomal localization. The karyotypic form A occurs in the distribution center of the type locality of A. paranae, in the proximities of the town of Castro (Tibagi basin), and may have reached the headwaters of the Ribeira River by the breakdown of geographical barriers. The karyotypic forms B and C are sympatric and syntopic, occurring solely in the Jaguariaíva River basin. Our hypothesis is that the karyotypic form A corresponds to the species A. paranae and forms B and C correspond to other species of the A. scabripinnis complex.

In order to refine the measure of karyotype asymmetry a new chromosomal parameter of dispersion index is proposed that has the potential to decipher even the minor karyotypic variations, thus permitting further evolutionary gradations to the karyotype asymmetry classes of Stebbins. The higher the dispersion index, the more specialized would be the karyotype. The dispersion index takes into account the variance lor gradual change in chromosome size within a complement vis-a-vis variance for the position of centromere in a karyotypic totality. The dispersion index is calculated as the proportionate measure of centromeric gradient to the coefficient of variation for chromosome length; wherin centromeric gradient = length of median short arm – length of median chromosome. Thus, the three most important karyotypic criteria viz., differences in: absolute chromosome size, position of centromere and relative chromosome size, are all covered in the proposed parameter. The effectiveness of dispersion index has been tested on a plant taxa, Papaver L., where karyomorphological details, nuclear DNA content, and morphotaxonomic parameters have been amply elucidated from an evolutionary stand point. It is hoped that dispersion index would find immense utility in delimiting species interrelationships particularly in the closely related taxa, when applied in conjunction with other systematic parameters.
